Restoring lipid homeostasis to redefine Alzheimer’s disease

MCM6 is developing disease-modifying treatments for Alzheimer’s disease for patients who carry APOE4, the largest genetic risk group.

We aim to restore lipid homeostasis using stem cell-based technology and a 3D multicellular brain iPS cell platform to identify drug targets, to optimize leads, and to conduct preclinical and clinical studies.

Science

The APOE4 isoform impairs glial lipid efflux and neuron–glia trafficking of cholesterol and other lipids, leading to aberrant lipid accumulation that initiates disease pathology.

Currently approved therapeutics target amyloid, tau, and synaptic transmission but not lipid homeostasis.

MCM6’s specific aim is to focus on this therapeutic gap by targeting lipid metabolism and restoring lipid homeostasis using stem cell–based technology and validated platforms.

Team

The team combines drug-discovery experience with expertise in APOE4 biology. Our goal is to target mechanisms of APOE4 cerebrovascular dysfunction and amyloid accumulation in Alzheimer’s disease.

The founders have experience in advanced programs spanning from target identification through lead optimization toward preclinical candidates.

The underlying science originates from the Blanchard Lab at Mount Sinai, and MCM6 is actively collaborating to advance into future IND-enabling studies.

Our story

In 1906, Dr. Alois Alzheimer described the neuropathological hallmarks of the disease that now bears his name. In Roman numerals, 1906 is MCMVI — the origin of the name MCM6.

Founded in recognition of that milestone, MCM6 is developing therapeutics to restore lipid homeostasis in APOE4 carriers, the largest genetic risk group for Alzheimer’s disease.

Pipeline

MCM6 translates biological knowledge of APOE4 lipid biology into druggable targets. Validation and lead optimization are underway. Next steps include identifying preclinical candidates and conducting IND-enabling studies.

Platform Capabilities

Established HCS (High-content screening) using human APOE4 iPSC-derived 2D neuronal cells and validated the leads with 3-D miBRAIN 3D multi-cell-based platform, ready for preclinical studies.

Current Progress

Target validation and lead optimization are underway, supported by NIA/NIH STTR Phase 1 and Phase 2 grant funding.

Next Milestones

Selection of preclinical candidates, IND-enabling studies and strategic partnerships to accelerate clinical development. MCM6 is advancing a portfolio of candidates with the potential to become first-in-class disease-modifying therapies for APOE4-associated Alzheimer’s disease.
